Nicotinamide adenine dinucleotide (NAD) is a well-known coenzyme found in all living cells that plays an important role in energy metabolism.NAD exists as an oxidized form (NAD + ) and a reduced form (NADH). The NAD + /NADH redox couple is essential for maintaining cellular redox homeostasis and for modulating numerous biological events, including cellular metabolism. Prolonged disequilibrium of NAD + /NADH balance disturbs the physiological functions, resulting in several diseases like metabolic diseases, neurodegeneration disorders or cancer.

 

In addition to its vital role as a coenzyme in energy metabolism, NAD +serves as substrate for three classes of enzymes: sirtuin family deacetylases, poly(ADP)-ribosyl polymerases and cADP-ribose synthases.

분석 원리 (Assay Principles)

The BQC NAD + /NADH Assay Kit is based on an enzymatic cycling reaction in which NAD + is reduced to NADH. The NADH present in the sample and the newly formed NADH is then oxidized by an electron intermediate acceptor resulting in the reduction of a tetrazolium salt to formazan.Formazan is a highly colored compound that shows a maximum absorption at 570 nm. The amount of formazan generated is proportional to the total amount of NAD (NAD + + NADH) in the sample.

 

The native NADH can be determined alone by heating the sample (thermal degradation of NAD + ) before the assay.
